Measurements are conducted with Leiz Orthoplan microscope and images are captured with peltier-cooled 12-bit CCD-camera (Fig. 1). Calibration, image capturing and required calculations are fully controlled with computer. The calibration is conducted with neutral density filter set.<\/p>\n\n\n\n<figure class=\"wp-block-image aligncenter size-full\"><img loading=\"lazy\" decoding=\"async\" width=\"411\" height=\"308\" src=\"https:\/\/sites.uef.fi\/biophysics\/wp-content\/uploads\/sites\/87\/2021\/09\/dd.jpg\" alt=\"\" class=\"wp-image-2368\" srcset=\"https:\/\/sites.uef.fi\/biophysics\/wp-content\/uploads\/sites\/87\/2021\/09\/dd.jpg 411w, https:\/\/sites.uef.fi\/biophysics\/wp-content\/uploads\/sites\/87\/2021\/09\/dd-300x225.jpg 300w\" sizes=\"auto, (max-width: 411px) 100vw, 411px\" \/><figcaption class=\"wp-element-caption\"><strong>Fig 1.<\/strong> Digital densitometry workstation.<\/figcaption><\/figure>\n\n\n\n<h4 class=\"wp-block-heading\">Polarized light microscopy<\/h4>\n\n\n\n<p>We have developed an enhanced polarized light microscope (ePLM), which is capable of measuring birefringence, orientation and anisotropy of any birefringent material. Polarization optics are replaced with precision grade components and placed to computer controlled rotation tables. The system is controlled with Macintosh G4 computer using IpLab 3.55 software (Fig. 2).<br><br><em>Features<\/em>:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>System can measure birefringence, orientation and anisotropy of collagen network<\/li>\n\n\n\n<li>Orientation independent measurement &#8211; all fibrils are measured with equal sensitivity<\/li>\n\n\n\n<li>Each pixel is individually calibrated for birefringence measurement &#8211; perfect calibrations (r<sup>2<\/sup>=1.000)<\/li>\n\n\n\n<li>Precise system &#8211; coefficient of variation 0.7%<\/li>\n\n\n\n<li>Wide dynamic range in detection &#8211; 12-bit camera<\/li>\n\n\n\n<li>Highly stable measurement system &#8211; 0.8% change over 5 hours in measured light intensity<\/li>\n<\/ul>\n\n\n\n<figure class=\"wp-block-image aligncenter size-full is-resized\"><img loading=\"lazy\" decoding=\"async\" width=\"411\" height=\"308\" src=\"https:\/\/sites.uef.fi\/biophysics\/wp-content\/uploads\/sites\/87\/2021\/09\/polarization.jpg\" alt=\"\" class=\"wp-image-2374\" style=\"width:445px;height:auto\" srcset=\"https:\/\/sites.uef.fi\/biophysics\/wp-content\/uploads\/sites\/87\/2021\/09\/polarization.jpg 411w, https:\/\/sites.uef.fi\/biophysics\/wp-content\/uploads\/sites\/87\/2021\/09\/polarization-300x225.jpg 300w\" sizes=\"auto, (max-width: 411px) 100vw, 411px\" \/><figcaption class=\"wp-element-caption\"><strong>Fig 2.<\/strong> Enhanced polarized light microscope (ePLM).<\/figcaption><\/figure>\n\n\n\n<h4 class=\"wp-block-heading\">Electron microscopy<\/h4>\n\n\n\n<p>We have Transmission Electron Microscope (TEM) and Scanning Electron Microscope (SEM) devices at our disposal at SIB-Labs at the University of Eastern Finland.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\">Confocal microscopy<\/h4>\n\n\n\n<p>We have a confocal laser scanning microscope at our disposal at <a href=\"https:\/\/www.uef.fi\/en\/unit\/institute-of-biomedicine\" target=\"_blank\" rel=\"noreferrer noopener\"><strong>Institute of Biomedicine<\/strong><\/a> at the University of Eastern Finland.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Optical density measurements We use digital densitometry (DD) to measure proteoglycan (PG) concentration of Safranin-O stained histological sections.
