WP1. UCNPs Synthesis, structural characterization, functionalization and biosafety tests.

Objectives:

Synthesis of FLUCNPs, FNDs, and GQDs suitable for the imaging of the biological tissues and cells.

Task 1.1. Synthesis of the FLUCNPs (INRS, UEF, ULM, UW, FTMC, UniSa, Platformina), M1-M36.

Task 1.2. Synthesis of the FNDs (ULM, UEF, UW, FTMC, UniSa, Platformina, INRS, Adamas), M1-M36.

Task 1.3. Fabrication of rare-earth ions doped GQDs (UEF, FTMC, ULM, Platformina), M1-M36.

D1.1. Protocol and technology of UCNPs synthesis. (UEF, M18).

D1.2. Protocol and technology of optimized synthesis of UCNPs selected for FLORIN super-resolving imaging scheme. (UEF, M42).

WP2. Spectroscopic characterization of UCNP. Experimental validation of UCNP biosensing properties.

Objectives:

UCNPs spectroscopic characterization for further optimization of their synthesis and functionalization routes, biosensing tests, demonstration of Quantum thermometry.

Task 2.1. Spectroscopic characterization (FTMC, UEF, ULM, UW, INRS, Adamas), M1-M40.

Task 2.2. Biosensing. Nanothermometry (ULM, UEF, FTMC, UW, Platformina), M3-M44.

Task 2.3. Comparative analysis of upconverting /emission performance of UCNPs (FTMC, INRS, ULM, UW, Atomicus, UniSa), M6-M44.

Deliverables:

D2.1. Spectroscopic characterization of first series of UCNPs. (ULM, M18).

D2.2. Report on biosensing and quantum nanothermometry. (ULM, M44).

WP3. Multiphysics modeling of UCNP emission and sensing properties.

Objectives:

Theory of the bioimaging based on FLUCNPs, FNDs, and GQDs with a focus on stochastic emission regime; modelling of the neuron cell electrophysiological response on the UCNPs emission.

Task 3.1. Stochastic emission regime modelling (Atomicus, UniSa, INRS, UW, UEF), M1-M24.

Task 3.2. Theoretical description of imaging and object inference method (ULM, UW, Atomicus, UEF, UniSa, Adamas), M6-M30.

Task 3.3. Finite element method modelling of a neuron under external forces. (UniSa, FTMC, UEF), M1-M30.

Deliverables:

D3.1. Data sheet of the most important parameters and KPIs based on theory and verified neuron model (ULM, M30).

WP4. SOFISM / Q-ISM with UCNPs and AI based data and image processing.

Objectives:

Implementation of the SOFISM / Q-ISM imaging schemes with produced UCNPs and biological samples.

Task 4.1. Development of SOFISM and QISM techniques with biological samples (UW, UEF, INRS, Adamas, FTMC, Platformina), M6-M24.

Task 4.2. AI based data processing (Atomicus, ULM, UEF, UniSa), M12-M30.

Deliverables:

D4.1. FLORIN device prototype (UW, M24).

D4.2. AI based data processing software ready for practical bioimaging (ATOMICUS, M30).

WP5. Practical bioimaging. Optogenetics. Proof of concept experiments.

Objectives:

Demonstration of the super-resolution bioimaging and biosensing using fabricated nanoparticles.

Task 5.1. Preparation and imaging of neuron cultures (UEF, UW, UniSa, Platformina, Adamas, IRNS), M12-M36.

Task 5.2. Imaging and Optogenetics (UW, UEF, FTMC, Platformina, ULM, Adamas, INRS), M12-M48.

Deliverables:

D5.1. SOFISM/Q-ISM approach tested with AD affected cell samples (UW, M24).

D5.2. Optogenetics testing and validation of neuronal- and glia-specific outcome measures related to nanoparticles in in vitro disease models of AD. (UEF, M48).

WP6. Training, networking, dissemination, communication.

Objectives:

WP6 focuses on the training, broad dissemination and communication of FLORIN project results and networking activities that will be carried out continuously. The main objectives are: (1) effective and sustainable dissemination of knowledge among and beyond the members of the consortium; (2) coordination of knowledge management and other innovation-related activities; (3) coordination of workshops and training activities within the project; (4) coordination of communication and outreach activities to public; (5) promotion of international and inter-sectoral long-lasting collaborations.

Task 6.1. Plan of Exploitation and Dissemination of project results, M1-M48, UEF, all partners.

Task 6.2. Website, M1-M48, UEF with participation of all partners on the day-by-day basis.

Objectives:
