Microglia function as sentinels of the central nervous system and take care of clearing and processing this fibrillar waste. The BBB is mainly formed by brain endothelial cells (ECs), pericytes and astrocytes that together with neurons and microglia present a functional unit called the neurovascular unit. Dysfunction of the BBB is recognized in several neurodegenerative diseases including Parkinson\u00b4s disease, but the mechanisms behind this are not fully known. Advances in human iPSC and organ-on-chip technology can provide more representative platforms to model and study the BBB function in a more <em>in vivo<\/em>-like context.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><a href=\"https:\/\/sites.uef.fi\/human-brain-disease-modelling-group\/lewy-bodies-and-dopaminergic-neurons\">Early Lewy body inclusions and genetic screening<\/a><\/h2>\n\n\n\n<figure class=\"wp-block-image size-full is-resized\"><img loading=\"lazy\" decoding=\"async\" width=\"325\" height=\"242\" src=\"https:\/\/sites.uef.fi\/human-brain-disease-modelling-group\/wp-content\/uploads\/sites\/286\/2021\/06\/dopaminergic.jpg\" alt=\"\" class=\"wp-image-1922\" style=\"aspect-ratio:1.3429752066115703;width:810px;height:auto\" srcset=\"https:\/\/sites.uef.fi\/human-brain-disease-modelling-group\/wp-content\/uploads\/sites\/286\/2021\/06\/dopaminergic.jpg 325w, https:\/\/sites.uef.fi\/human-brain-disease-modelling-group\/wp-content\/uploads\/sites\/286\/2021\/06\/dopaminergic-300x223.jpg 300w\" sizes=\"auto, (max-width: 325px) 100vw, 325px\" \/><\/figure>\n\n\n\n<p>Lewy bodies are present in multiple neurogenerative diseases, such as Parkinson&#8217;s disease, Alzheimers disease and Lewy body dementia. \u03b1-synuclein is one of the key molecules found inside of Lewy bodies and it is routinely used to model \u03b1-synuclein pathology and Lewy body-like inclusions in neurodegenerative disease models. In the central nervous system, they maintain the function of blood-brain barrier (BBB), regulate inflammation, and take up and degrade different proteins. Recent data has indicated that microglia may contribute to disease by producing an overactive proinflammatory response and having a dysfunction in their ability to phagocytose pathological proteins, including&nbsp;\u03b1-synuclein. This project aims to unveil the role of gut changes in Parkinson\u2019s disease and identify potential therapeutic targets by creating <em>in vitro<\/em> models of the microbiota-gut-brain-axis. The project will utilize human iPSC-derived enteric and dopaminergic neurons together with intestinal organoids to study the complex interplay between microbiota and the brain<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><a href=\"https:\/\/sites.uef.fi\/human-brain-disease-modelling-group\/the-impact-of-environment-polluting-agents-in-parkinsons-disease\/\">The impact of environment polluting agents in Parkinson&#8217;s disease<\/a><\/h2>\n\n\n\n<figure class=\"wp-block-image size-full\"><img loading=\"lazy\" decoding=\"async\" width=\"792\" height=\"368\" src=\"https:\/\/sites.uef.fi\/human-brain-disease-modelling-group\/wp-content\/uploads\/sites\/286\/2025\/02\/image.jpeg\" alt=\"\" class=\"wp-image-4190\" srcset=\"https:\/\/sites.uef.fi\/human-brain-disease-modelling-group\/wp-content\/uploads\/sites\/286\/2025\/02\/image.jpeg 792w, https:\/\/sites.uef.fi\/human-brain-disease-modelling-group\/wp-content\/uploads\/sites\/286\/2025\/02\/image-300x139.jpeg 300w, https:\/\/sites.uef.fi\/human-brain-disease-modelling-group\/wp-content\/uploads\/sites\/286\/2025\/02\/image-768x357.jpeg 768w\" sizes=\"auto, (max-width: 792px) 100vw, 792px\" \/><\/figure>\n\n\n\n<p>Most of the patients experience the condition spontaneously, suggesting a significant role of environmental factors in disease progression. This project aims to provide new insights into the complex aetiology of PD by utilizing human dopaminergic neurons and a 3D organoid model.
